Yes, all three buttons (mouse 1, 2, and the scroll wheel) are fully programmable via the SteelSeries Engine (this does not include the fourth CPI button which is used to toggle CPI).
The buttons can be assigned either to keyboard keys or mouse functions, which includes the capacity for multiple click macros.
